現在、世界の海には 250 000 トンを超えるプラスチックごみが浮かんでいます。太平洋は北半球で最も汚染された水域と評価され、南半球ではインド洋が太平洋と大西洋を合わせた以上に汚染されていることが分かりました。同時に、太平洋は地球上でプラスチックごみに最も汚染された海とされています。プラスチック汚染は、卓越風や海流に運ばれて世界の海を移動します。北半球では、数年にわたる長期的な海水とごみの移動によって、海盆の中央にプラスチックごみが集まることが示されています。南半球のすべての海でも、同様の傾向が確認されています。驚くことに、南半球の海で測定されたプラスチックの総量は、北半球と同じ数量の範囲にあります。北半球には南半球よりはるかに多くの流入源があることを考えると、意外な結果です。これは、プラスチック製品が以前の想定よりも容易に海流間や半球間を移動し、再分布することを意味します。
しかし、浮遊するマイクロプラスチックの最終的な行き先は海面ではありません。海面には予想よりはるかに少ないマイクロプラスチックしかないという観察は、プラスチックごみを水面から取り除く何らかの過程があることを示しています。そこには紫外線による分解、生分解、生物による摂取、生物の付着による浮力の低下と海底への沈降、海岸への打ち上げが含まれます。
プラスチックごみは、物理化学的な非生物的過程、または生物的な生分解によって分解されます。機械的な物理力によるポリマー材料の破壊は通常、最初の段階と考えられ、どの分解過程でも非常に重要な役割を果たします。環境中のプラスチックの分解は、光分解、加水分解、熱酸化分解、生分解という四つの主な仕組みの一つ、または組み合わせによって起こります。海洋環境など自然条件下では、低圧法ポリエチレン、高圧法ポリエチレン、ポリプロピレン(LDPE、HDPE、PP)などの一般的なプラスチックの分解は、主に UV-B の作用による光分解から始まり、熱酸化、そしてより小さな程度で水による分解、つまり加水分解へと進みます。
ウッズホール海洋研究所の科学者が発表した新しい研究は、世界で最も普及しているプラスチックの一つであるポリスチレン(PS)が、太陽光によって、従来考えられていた数千年ではなく数十年から数百年で分解され得ることを示しています。ポリスチレンの部分的な分解は、種類によってわずか 10–50 年で起こります。完全な光化学的酸化は 300–450 年で起こり得ます。太陽光はプラスチックを物理的に壊すだけでなく、溶存有機炭素と、大気に移る少量の二酸化炭素へと化学的に分解します。
分解過程は、プラスチックをより小さな破片に砕く断片化と、ポリマーの分子量の低下を引き起こします。その後、低分子量の物質は微生物に代謝されます。いずれにしても、全体の過程は通常とても遅く、他の生物がプラスチックポリマーを完全に分解するには 50 年以上かかる場合があります。
多くの複雑な天然・合成化合物は、単独の菌株ではなく微生物の共同体によって生分解されます。これはおそらく、個々の生物の代謝能力が限られているためです。そのため、現代社会が直面する主要な環境問題の一つであるプラスチックごみは、異なる生物群が協力することで、より効率よく分解できると考えられます。プラスチックの生分解とは主に、嫌気性微生物が空気のない状態でポリマー材料をバイオガスとバイオマスに変えることを指します。これらの微生物は、成長の炭素源としてポリマーを効率よく利用できます。しかし、プラスチック自体の性質と特有の物理化学的性質が生分解を妨げ、成長の基質として適さないものにしています。
プラスチックごみの生分解は、酵素によってポリマー鎖がオリゴマー、二量体、単量体などの低分子量化合物へ切断されることから始まります。酵素がポリマーに結合し、水による加水分解を触媒する、つまり速めることで進みます。最後に、これらの低分子化合物は二酸化炭素と水へ変わります。細菌、菌類、藻類は、プラスチックポリマーを分解できる有効な生物として報告されています。
菌類はポリマー材料の分解で中心的な役割を担います。菌糸体は表面から内部へ効率よく入り込み、できる限り多くの基質を分解できます。また、デポリメラーゼなどの細胞外酵素を分泌し、ポリマー基質をオリゴマー、二量体、単量体という低分子量の断片へ切断できます。単量体は菌類に取り込まれ、同化されるか、細胞内の酵素系によって二酸化炭素と水へ変わります。この観点から、白色腐朽菌と褐色腐朽菌は、プラスチックポリマーを効率よく分解する菌としてよく挙げられます。
同様に、細菌の菌株も汚染された水や土壌の中でプラスチックのポリマーを分解できます。複数の研究では、特化した細菌によるプラスチックの生分解が、汚染された生態系のバイオレメディエーションに有望な方法となることが報告されています。Pseudomonas spp.、Bacillus spp.、Streptomyces spp. などの菌株は、さまざまなプラスチックポリマーを高い効率で分解します。いずれにしても、菌類によるプラスチックの分解速度は、細菌の菌株で得られる速度を上回ります。一方で、安定した条件をより必要とする菌類に比べ、細菌は培養しやすく、ポリマー材料の分解に利用しやすいと報告する著者もいます。
これまで、藻類がプラスチックごみ汚染を緩和する能力を報告した研究はわずかです。観察されたのは、成長に欠かせない太陽光、栄養、水などの環境要因があることで、糸状藻類がプラスチックごみの表面に定着できるということでした。Bacillariophyceae、Chlorophyceae、Cyanophyceae に属する無害で無毒のいくつかの藻類は、ポリエチレン表面に生育し、池、湖、排水などの汚染された水域で藻類のバイオフィルムを形成できると分かっています。この点について著者らは、分離しやすく成長の速い Phormidium lucidum と Oscillatoria subbrevis、すなわち淡水の無毒なシアノバクテリアが、PE 表面にコロニーを作り、前処理なしで HDPE を効率よく分解できると報告しています。
さらに、PETase 酵素の生産に使われる Ideonella sakaiensis や Pestalotiopsis microspore などの細菌・菌類は、プラスチックごみの大部分が蓄積する海洋環境にはうまく適応できません。珪藻 Phaeodactylum tricornutum を微生物工場として利用し、PET と共重合体 PETG(グリコール変性ポリエチレンテレフタレート)に活性を示す改良型 PETase を生産することに成功しています。
ポリマーの生分解は、スーパーワーム Zophobas atratus、ノシメマダラメイガ Plodia interpunctella、ハチノスツヅリガ Galleria mellonella、ミールワームの幼虫 Tenebrio molitor、ウスグロツヅリガ Achroia grisella、陸生のカタツムリ(Achatina fulica)、シロアリ、その他の無脊椎動物でも確認されています。プラスチックポリマーを分解するこの優れた働きは、通常、腸内の共生微生物によるものとされています。いくつかの無脊椎動物が木材やその他のポリマー化合物を食べられるのも、それらの微生物が関わっているからです。
プラスチックは独特の性質によって広く使われるようになり、生活に最も欠かせない材料の一つと考えられています。しかし、プラスチックごみの有害な影響は、使用量と環境への蓄積量に比例します。そのため、プラスチック材料の性質は、微生物による分解だけでなく、表面への定着にも大きな課題となります。多くの研究者が、複数の微生物や無脊椎動物のプラスチック生分解への関与を指摘しており、材料の生分解過程でそれらが基本的な役割を担うことが示されています。
こうしたことから、専門の科学者は、プラスチックごみの物理的・化学的な前処理を、最初の重要な段階として強調しています。それはしばしば、微生物がプラスチックポリマーを分解するための前提となります。微生物、藻類、プラスチックを食べる昆虫、その他の無脊椎動物は、将来のプラスチック生分解で重要な役割を果たす可能性があります。研究は、環境中へのプラスチックごみの蓄積を克服するために、環境にやさしく、効率的で安価な処理・分解技術の開発を提案しています。
